Why was Khorana's chemical synthesis of genes important?
The chemical synthesis of genes was paramount because it allowed us to move beyond observation to active construction and proof. To truly understand the function of heredity, we needed to build the molecules that carry that information. By synthesizing specific sequences of nucleotides, we could control the 'blueprint.' This enabled us to test hypotheses about gene function and to create biologically active DNA molecules. Our work on the total synthesis of a tyrosine suppressor tRNA gene demonstrated that we could assemble a functional DNA molecule from its basic chemical components, providing irrefutable evidence for the chemical basis of genetic information transfer and function. In biology, the chemistry must be right.
